| Comments | The tet response element (TRE) is placed upstream of the human cdk6 cDNA in this Ad5 backbone adenovirus. The cdk6 construct also contains a C-terminal HA tag. When human islets are co-transduced with the adenovirus that uses the CMV-driven tet-transactivator (TTA), the cdk6 expression assessed by both immunohistochemistry and immunoblot, using either cdk6 or HA antibodies, is abundant, and increases in a dose-related manner over 72 hours with doses beginning at 0.1 ug/ml, and increasing to 1 µg/ml. This construct was generated using pACCMVpLpA & pJM17 in HEK293 cells. |
