| Notes: | The monoclonal antibody produced by hybridoma DMBC0 8-E10 reacts with a cell surface molecule on mouse ES-derived definitive endoderm. The antibody reacts with definitive endoderm derived from ES cells differentiated for 6D with activin (Foxa3 positive cells), and does not react with ES cells differentiated under conditions giving rise to mesoderm or ectoderm. The monoclonal antibody has also been found to react with cells in the developing mouse embryo. This hybridoma was developed by the Keller, Streeter, and Grompe Laboratories. |

Description: A standard two-step staining strategy can be used with this rat monoclonal antibody. Incubate test cells with supernatant (30 min on ice). Wash cells 1X with PBS containing 2% FBS. Resuspend cells in a fluorochrome-conjugated secondary anti-rat Ig (H+L). The secondary antibody used in the figures shown here was an APC-donkey anti-rat IgG (H+L) (Jackson ImmunoResearch Laboratories, Inc.; diluted 1:200). Wash cells again in PBS and evaluate.
If additional staining with a conjugated rat antibodies is desired, block any anti-rat binding sites with rat IgG (50 ug/mL for 10 min), then add titrated conjugated rat antibody. Protocols: |
