ESPRESSO (ES cells for PancReas ExpreSSiOn) Project
The ESPRESSO (ES cells for PancReas ExpresSSiOn) project was designed to collect all existing targeted ES cells created by Beta Cell Biology Consortium (BCBC) investigators. Available here are germline-competent mESC lines and detailed information on the vectors used to generate these constructs with.

Recombineering, Gene Targeting and RMCE Vectors
These recombineering vectors can be used for any LCA construction and are available from www.addgene.org (specific links are provided below). In addition, there are two protocols associated with each of these vectors:
| Vector | Description | Genbank File | Request |
|---|---|---|---|
| pBS.DT-A | A gap-repair vector designed for retrieving the targeted region of BAC clones to construct gene targeting vectors using a recombineering assembly strategy. The plasmid contains a diphtheria toxin selection gene, DT-A. | pBS.DT-A.gb |
|
| pBS.L66.2272 | This vector is modified from pBS KS+. A lox66 site, flanked by Sac I and Not I, and a lox2272 site, flanked by Xho I and Kpn I, were inserted into the same sites of pBluescript KS+. | pBS.L66.2272.gb |
|
| phygro66.2272.F | This vector is modified based on pBS.L66.2272. A FRT2.hygro Not I fragment from pFRT2.hygro was inserted into the Not I site in 5’->3’ orientation. | phygro66.2272.F.gb |
|
| pLCA.71.2272.NTK+Xho | This vector was designed for assembling gene targeting constructs using the recombineering approach and the Cre-lox system. The vector contains a puromycin-Dthymidine kinase fusion gene driven by the mouse phosphoglycerol kinase promoter (pUDTK) and a neomycin resistant gene driven by the bacterial EM7 promoter (EM7neo). These selectable markers are flanked by lox71 and lox2272 sites (Cre-recombinase recognition sequences) in a pBluescript KS+ backbone with a modified multi-cloning region. | pLCA.71.2272.NTK+Xho.gb |
