Our principal objective is to identify and characterize transcription factors essential in pancreatic islet development and function, with a special focus on those of the insulin gene. Pancreatic beta-cell-type-specific and glucose-induced insulin expression appears to be principally regulated by the PDX-1, MafA, and NeuroD1/BETA2 transcription factors. Individuals heterozygous for an inactivating mutation in PDX-1 and BETA2 are diabetic, presumably due to beta cells no longer producing sufficient insulin to meet the body''s needs.
